Results

A 3D imaging system captures fluorescence emissions from samples and relays them via an intermediate image plane via a tube lens (TL, f = 200 mm, ITL200 from Thorlabs). An intermediate image was then spatially transformed utilizing a focusing mirror (FM, 160mm focal length, S600-f28 from RPC Photonics) into a 5×5 sub-image array. Sensor events are generated when intensity variations exceed a predefined threshold, providing information such as latency, timestamp, and the directionality of intensity changes. Polarity-sorted event streams are integrated within an accumulation time selected by the user to reconstruct temporally continuous frames, as shown in Fig. 1b with signal-bearing beads moving linearly and blinking as expected (as evidenced by bead motion in linear frames).

To measure the performance of EventLFM, we conducted a side-by-side comparison between it and conventional scanning-based widefield microscopy and cutting-edge single-shot 3D light field microscopy (Fourier LFM). Specifically, we evaluated resolution across various velocity ranges by measuring full width at half maximum (FWHM) profiles reconstructed from bead intensity profiles; Figure 2 displays representative time-surface frames from both systems along with their 3D depth color-coded reconstructions.

EventLFM proved itself adept at reconstructing bead intensity profiles with consistent intensity across a broad velocity range, boasting excellent axial resolution at high frame rates and providing high frame rates without compromising spatial or temporal resolution. Furthermore, EventLFM demonstrated its capability of detecting fast dynamic signals despite high noise levels without impairing either spatial or temporal resolution.

EventLFM proved its ability to track and extract neuronal spike trains using a 3D phantom embedded with fluorescent beads, with matching reconstructions against ground truth stimuli. EventLFM successfully detected dynamic features of neuronal signals even in the presence of dynamic effects such as LED pulse sequences; this paves the way for more advanced computational algorithms that utilize event-driven measurements for extracting physically meaningful signals from sparse data in deep scattering media environments.
